چکیده

Recommended Citation Wongrakpanich, Amaraporn. "The development of cationic polymers for non-viral gene delivery system." PhD (Doctor of Philosophy) thesis, ii To my parents iii ACKNOWLEDGMENTS My deep gratitude goes first to my advisor, Dr. Aliasger K. Salem for giving me the opportunity to do research in his lab and teaching me how to design and carryout experiments independently. His ability to always recognize the positive side of any situation allowed me to have confidence in my work and encouraged me to stay motivated. This helped a lot in my research as things don't always go as expected. I hope to repay his gratitude towards me and show him how much I learned during the time spent in his lab. I look forward to our collaboration in the future. I would also like to show appreciation to my thesis and comprehensive exam committee, including Dr. Assem for their insightful suggestions and discussion during the progress of this work that has made me a better researcher. I would like to thank Dr. Douglas Flanagan, from whom I took two courses that enhanced my ability to not only learn and think on my own but to really understand difficult concepts rather than just using memorization alone. Joiner for teaching and guiding me with their expertise in in vivo pulmonary delivery methods, High-Content Screening and mitochondria model in heart diseases, respectively. I thank Kathy Walters and Chantal Allamargot of Central Microscopy Research Facility for teaching me useful microscopy techniques. Also, the graduate students who worked closely with me; and others that are not mentioned here. iv All of this work would not have been possible without my labmates. I would like to thank Dr. Sean Geary (who helped me tremendously with writing, taught me to think like a scientist and who is a great friend), Vijaya Joshi (who taught me everything in the lab from pipetting, handling biohazardous waste to making complicated fancy particles), Chitphet, Cristina Maria de Barros and Emad Wafa with the wonderful scientific discussions and helping me with anything I needed. I will never forget the great times I had during lab gatherings, chit chatting during group lunches, sipping coffee on the deck or even fighting for space in the cell culture hood. I am looking forward to reading about your guys' work in the big journals later! I also want to acknowledge Brenda Zobeck for helping me with administrative work …

Investigating lipopolymers based on polyethylenimine and nanoliposome for gene delivery to prostate cancer (PC3) cell line

Background: Non-viral Nano carriers such as liposomes and cationic polymers based on engineered properties are regarded in gene delivery field. Although these carriers do not have weaknesses of viral vectors, but they are less efficient than viruses and they still need to be improved as favorable gene delivery carriers. Amongst non-viral carriers, cationic liposomes have been proposed for clini...

Non-biological gene carriers designed for overcoming the major extra- and intracellular hurdles in gene delivery, an updated review

Gene therapy as a modern therapeutic approach has not yet advanced to a globally-approved therapeutic approach. Lack of adequate reliable gene delivery system seems to be one of the major reasons from the pharmaceutical biotechnology point of view. Main obstacles delaying successful application of human gene therapy are presented in this review. The unique advantages of non-biological gene carr...

Evaluation of Cell Penetrating Peptide Delivery System on HPV16E7 Expression in Three Types of Cell Line

Background: The poor permeability of the plasma and nuclear membranes to DNA plasmids are two major barriers for the development of these therapeutic molecules. Therefore, success in gene therapy approaches depends on the development of efficient and safe non-viral delivery systems. Objectives: The aim of this study was to investigate the in vitro delivery of plasmid DNA encoding HPV16 E7 gene...

Formulation of a therapeutic cationic liposome-siRNA complex for development to fight osteosarcoma

Introdution: Cationic liposomes have been presented for gene delivery as an alternative vector instead of viral vectors. A major challenge associated with siRNA delivery is the instability of liposomes, which is still a serious problem. The aim of this study was to provide an appropriate formulation to overcome this instability. Methods: In the present study (Scientific-Fundamental, Experiment...

Barriers and recent advances in non-viral vectors targeting the lungs for cystic fibrosis gene therapy

Cystic fibrosis (CF) is an autosomal recessive disorder caused by mutations in CFTR genes that affect chloride ion channel. The CF is a good nominee for gene therapy as the asymptomatic carriers are phenotypically normal, and the desired cells are accessible for vector delivery. Gene therapy shows promising effects involving the correction of gene or replacement of the mutant gene with the func...

Cellular Uptake Mechanism of Non-Viral Gene Delivery and Means for Improving Transfection Efficiency

Non-viral delivery systems usually include mechanical, electrical, and chemical methods. Cationic liposomes and cationic polymers are two typical classes of non-viral vectors. Com‐ pared with viral vectors, non-viral ones are considered promising vehicles for gene therapy because of their low toxicity, biocompatibility, and controllability [1, 2], although their low efficacy limits their applic...
